Biological Sample Preparation for SEM Imaging of Porcine Retina

چکیده

Introduction Sample preparation is a critical step in scanning electron microscopy (SEM) imaging. This is especially true for biological samples because of charge build-up and sensitivity to vacuum and electron beam damage. In terms of ultrastructure imaging, a variety of advancements in detectors and approaches have improved biological imaging such that fewer steps are required for sample preparation. However, the conventional approach incorporating osmium tetroxide fixing, ethanol dehydrating, critical-point drying, and coating still finds useful application. This paper evaluates three biological sample-preparation methodologies for imaging the ultrastructure of immature porcine retina. The three preparation methods examined are critical-point drying (CPD), hexamethyldisilazane (HMDS) dehydration, and direct imaging by environmental scanning electron microscopy (ESEM). Preparation methodologies were evaluated based on resulting image quality and reduced potential for artifacts.

Specimen Preparation for Electron Microscopy

In general the SEM sample preparation techniques are more or less similar to the metallographic sample preparation technique for optical microscopy, because both the microscopes reveals the surface topography of sample. In SEM a focussed beam of electrons scans over the specimen surface and the electrons emitted from the sample surface controls the brightness of the CRT spot which also scans th...

Application of Ionic Liquid on Biological Samples in Correlative Optical Microscopy and Scanning Electron Microscopy

Correlative optical microscopy and scanning electron microscopy (SEM) combines two techniques enabling the use both optical and electronic analysis techniques on the same sample. For example, optical techniques, such as fluorescence staining, fluorescence in situ hybridization (FISH) could in principle be used with SEM and energy dispersive spectroscopy (EDS) [1]. Optical microscopy provides ef...

Quantitating morphological changes in biological samples during scanning electron microscopy sample preparation with correlative super-resolution microscopy

Sample preparation is critical to biological electron microscopy (EM), and there have been continuous efforts on optimizing the procedures to best preserve structures of interest in the sample. However, a quantitative characterization of the morphological changes associated with each step in EM sample preparation is currently lacking. Using correlative EM and superresolution microscopy (SRM), w...

Cryo-scanning electron microscopy of plant samples without metal coating, utilizing bulk conductivity

A classical problem in scanning electron microscopy is the alteration of specimen surfaces due to sample preparation. Simplified techniques for sample preparation with minimized surface alteration are therefore desirable. For fresh (hydrated) biological material, cryo-SEM after preceding shock-freezing and metal-coating is favoured when the prevention of drying artifacts has priority. A detaile...

Surface damage induced by FIB milling and imaging of biological samples is controllable.

Focused ion beam (FIB) techniques are among the most important tools for the nanostructuring of surfaces. We used the FIB/SEM (scanning electron microscope) for milling and imaging of digestive gland cells. The aim of our study was to document the interactions of FIB with the surface of the biological sample during FIB investigation, to identify the classes of artifacts, and to test procedures ...
